Yes. You can test a motherboard without a CPU cooler but it’s not recommended.

If you’re just testing to see if the board is working you can temporarily place a metal screwdriver or other metal object across the top of the CPU to provide some basic cooling.

But be very careful not to touch any of the components on the bottom of the motherboard with the metal object.

If you do you could create a short circuit and damage the board.

If you’re testing to see if the board is overclocking properly you’ll need to install some kind of cooling system (either air or water-based system) in order to get accurate results.
